Blueno knows that if Lucci is ever forced to kill Kaku, then the strongest CP9 member will regret it. The regret will not stop him from doing so, but he will regret it. That's how Blueno knows that Lucci respects Kaku.
Jyabura knows that the only person Lucci respect is Kaku, because he didn't laugh even when seeing Kaku's ridiculous and square half-giraffe form. And to Jyabura, that's the surest way to know that the damn alley-cat respects the long nosed freak.
Fukurou has heard many rumours in his days, and he's spread even more. He's an expert at spying on his colleagues to find out about their personal lives and the day he found out that Kaku helps Lucci with Hattori when the bird gets sick or has to stay back at Enies Lobby when Lucci is on a mission, that's when Fukurou knew that Kaku respects Lucci and that Lucci trusts Kaku.
Kumadori thinks that it is wonderful to see the two strongest CP9 members interact. They can hold entire conversations through glances, and once, Kumadori swears that he saw Lucci give Kaku a smile, and a pleasant one at that. So Kumadori knows that they trust and respect each other.
Kalifa knows that men are stupid to the point of retardation, even most of her co-workers. Lucci has no feelings according to most people, he consists of nothing. Kaku is the same, despite his ability to simulate emotions. And what happens when nothing meets nothing? Well, nothing, except that the two nothings blend together to create a bigger nothing. That's how Kalifa knows, despite how everyone would deny or misinterpret it, that Kaku and Lucci must love each other. (The fact that she has caught them in bed on more than one occasion helps).
